ROLE OF CARVACROL IN PROLIFERATIVE CELL SIGNALLING AND ISCHEMIC STROKES: A DETAILED REVIEW

Özet

Bitki bazlı moleküller, antik çağlardan beri terapötik ajanlar olarak insan hastalıklarının tedavisinde önemli roller oynamıştır. Karvakrol (2-metil-5-izopropil fenol), biyoaktif bileşiklerin izoprenoid grubu arasında en çok çalışılanlar arasındadır ve farmasötik, gıda, biyoteknoloji endüstrileri ve diğerlerinde çeşitli uygulamalara sahiptir. Antimikrobiyal, antikanser, antispazmodik, immünomodülatör, antiinflamatuar, antioksidan ve mitojenik etkilerinin keşfi onu çok sayıda araştırma projesine konu etmiştir.  Bu çeşitli etkilerin altında yatan moleküler mekanizmalar halen devam eden araştırmaların konusudur ve bu konuda mevcut literatürler oldukça azdır. Bu derleme çalışması, hücrenin hayatta kalması (çoğalma) ve ölüm (apoptoz) konusundaki karvakrol-hücre sinyal yolu ilişkisi üzerine mevcut az sayıdaki literatürü ve temel ve translasyonel araştırmalar için gelecekte ortaya çıkan yönelimleri incelemeyi amaçladı. Carvacrol'ün, MAPK sinyal ailesinin üyeleri ERK1/2, p38, JNK ile ve  PI3K/AKT, IL6/STAT3, eNOS  gibi hücrenin hayatta kalması ve ölümü için gerekli olan önemli hücre sinyal yollarını aktive ettiği bilinmektedir. Mitokondriyal aracılı doza bağlı apoptoz ve sitoproteksiyon karvakrol etki mekanizmasının önemli bir özelliğidir. Karvakrol aynı zamanda siklin D1, siklin B1, CDK4, CDK6, pRb, vb hücre siklusunda anahtar rol oynayan önemli proteinlerin sentezin de düzenleyici olarak rol oynamaktadır.
